    A DHT-Based Discovery Service for the Internet of Things

    Current trends towards the Future Internet are envisaging the conception of novel services endowed with context-aware and autonomic capabilities to improve end users' quality of life. The Internet of Things paradigm is expected to contribute towards this ambitious vision by proposing models and mechanisms enabling the creation of networks of "smart things" on a large scale. It is widely recognized that efficient mechanisms for discovering available resources and capabilities are required to realize such vision. The contribution of this work consists in a novel discovery service for the Internet of Things. The proposed solution adopts a peer-to-peer approach for guaranteeing scalability, robustness, and easy maintenance of the overall system. While most existing peer-to-peer discovery services proposed for the IoT support solely exact match queries on a single attribute (i.e., the object identifier), our solution can handle multiattribute and range queries. We defined a layered approach by distinguishing three main aspects: multiattribute indexing, range query support, peer-to-peer routing. We chose to adopt an over-DHT indexing scheme to guarantee ease of design and implementation principles. We report on the implementation of a Proof of Concept in a dangerous goods monitoring scenario, and, finally, we discuss test results for structural properties and query performance evaluation

    A Service-Oriented Approach for Network-Centric Data Integration and Its Application to Maritime Surveillance

    Maritime-surveillance operators still demand for an integrated maritime picture better supporting international coordination for their operations, as looked for in the European area. In this area, many data-integration efforts have been interpreted in the past as the problem of designing, building and maintaining huge centralized repositories. Current research activities are instead leveraging service-oriented principles to achieve more flexible and network-centric solutions to systems and data integration. In this direction, this article reports on the design of a SOA platform, the Service and Application Integration (SAI) system, targeting novel approaches for legacy data and systems integration in the maritime surveillance domain. We have developed a proof-of-concept of the main system capabilities to assess feasibility of our approach and to evaluate how the SAI middleware architecture can fit application requirements for dynamic data search, aggregation and delivery in the distributed maritime domain

    Are there more galaxies than we see around high-zz quasars?

    Whether or not z6z \gtrsim 6 quasars lie in the most massive dark-matter halos of the Universe is still a subject of dispute. While most theoretical studies support this scenario, current observations yield discordant results when they probe the halo mass through the detection rate of quasar companion galaxies. Feedback processes from supermassive black holes and dust obscuration have been blamed for this discrepancy, but the impact of these effects is complex and far from being clearly understood. This paper aims to improve the interpretation of current far-infrared observations by taking into account the cosmological volume probed by the Atacama Large Millimeter/submillimeter Array Telescope and to explain the observational discrepancies. We statistically investigate the detection rate of quasar companions in current observations and verify if they match the expected distribution from various theoretical models, once convolved with the ALMA field-of-view, through the use of Monte Carlo simulations. We demonstrate that the telescope geometrical bias is fundamental and can alone explain the scatter in the number of detected satellite galaxies in different observations. We conclude that the resulting companion densities depend on the chosen galaxy distributions. According to our fiducial models, current data favour a density scenario where quasars lie in dark-matter halos of viral mass Mvir1012 MM_{\rm vir} \gtrsim 10^{12}~{\rm M_{\odot}}, in agreement with most theoretical studies. According to our analysis, each quasar has about 2 companion galaxies, with a [CII] luminosity L[CII]108 LL_{\rm [CII]} \gtrsim 10^8~{\rm L}_{\odot}, within a distance of about 1~Mpc from the quasar.Comment: 7 pages, 5 figures; accepted for publications in A&

    Biochemical properties and base excision repair complex formation of apurinic/apyrimidinic endonuclease from Pyrococcus furiosus

    Apurinic/apyrimidinic (AP) sites are the most frequently found mutagenic lesions in DNA, and they arise mainly from spontaneous base loss or modified base removal by damage-specific DNA glycosylases. AP sites are cleaved by AP endonucleases, and the resultant gaps in the DNA are repaired by DNA polymerase/DNA ligase reactions. We identified the gene product that is responsible for the AP endonuclease activity in the hyperthermophilic euryarchaeon, Pyrococcus furiosus. Furthermore, we detected the physical interaction between P. furiosus AP endonuclease (PfuAPE) and proliferating cell nuclear antigen (PCNA; PfuPCNA) by a pull-down assay and a surface plasmon resonance analysis. Interestingly, the associated 3′–5′ exonuclease activity, but not the AP endonuclease activity, of PfuAPE was stimulated by PfuPCNA. Immunoprecipitation experiments using the P. furiosus cell extracts supported the interaction between PfuAPE and PfuPCNA in the cells. This is the first report describing the physical and functional interactions between an archaeal AP endonuclease and PCNA. We also detected the ternary complex of PfuPCNA, PfuAPE and Pfu uracil-DNA glycosylase. This complex probably functions to enhance the repair of uracil-containing DNA in P. furiosus cells

    Functional capacity of XRCC1 protein variants identified in DNA repair-deficient Chinese hamster ovary cell lines and the human population

    XRCC1 operates as a scaffold protein in base excision repair, a pathway that copes with base and sugar damage in DNA. Studies using recombinant XRCC1 proteins revealed that: a C389Y substitution, responsible for the repair defects of the EM-C11 CHO cell line, caused protein instability; a V86R mutation abolished the interaction with POLβ, but did not disrupt the interactions with PARP-1, LIG3α and PCNA; and an E98K substitution, identified in EM-C12, reduced protein integrity, marginally destabilized the POLβ interaction, and slightly enhanced DNA binding. Two rare (P161L and Y576S) and two frequent (R194W and R399Q) amino acid population variants had little or no effect on XRCC1 protein stability or the interactions with POLβ, PARP-1, LIG3α, PCNA or DNA. One common population variant (R280H) had no pronounced effect on the interactions with POLβ, PARP-1, LIG3α and PCNA, but did reduce DNA-binding ability. When expressed in HeLa cells, the XRCC1 variants—excluding E98K, which was largely nucleolar, and C389Y, which exhibited reduced expression—exhibited normal nuclear distribution. Most of the protein variants, including the V86R POLβ-interaction mutant, displayed normal relocalization kinetics to/from sites of laser-induced DNA damage: except for E98K and C389Y, and the polymorphic variant R280H, which exhibited a slightly shorter retention time at DNA breaks

    Mortality and pulmonary complications in patients undergoing surgery with perioperative SARS-CoV-2 infection: an international cohort study

    Background: The impact of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) on postoperative recovery needs to be understood to inform clinical decision making during and after the COVID-19 pandemic. This study reports 30-day mortality and pulmonary complication rates in patients with perioperative SARS-CoV-2 infection. Methods: This international, multicentre, cohort study at 235 hospitals in 24 countries included all patients undergoing surgery who had SARS-CoV-2 infection confirmed within 7 days before or 30 days after surgery. The primary outcome measure was 30-day postoperative mortality and was assessed in all enrolled patients. The main secondary outcome measure was pulmonary complications, defined as pneumonia, acute respiratory distress syndrome, or unexpected postoperative ventilation. Findings: This analysis includes 1128 patients who had surgery between Jan 1 and March 31, 2020, of whom 835 (74·0%) had emergency surgery and 280 (24·8%) had elective surgery. SARS-CoV-2 infection was confirmed preoperatively in 294 (26·1%) patients. 30-day mortality was 23·8% (268 of 1128). Pulmonary complications occurred in 577 (51·2%) of 1128 patients; 30-day mortality in these patients was 38·0% (219 of 577), accounting for 81·7% (219 of 268) of all deaths. In adjusted analyses, 30-day mortality was associated with male sex (odds ratio 1·75 [95% CI 1·28–2·40], p\textless0·0001), age 70 years or older versus younger than 70 years (2·30 [1·65–3·22], p\textless0·0001), American Society of Anesthesiologists grades 3–5 versus grades 1–2 (2·35 [1·57–3·53], p\textless0·0001), malignant versus benign or obstetric diagnosis (1·55 [1·01–2·39], p=0·046), emergency versus elective surgery (1·67 [1·06–2·63], p=0·026), and major versus minor surgery (1·52 [1·01–2·31], p=0·047). Interpretation: Postoperative pulmonary complications occur in half of patients with perioperative SARS-CoV-2 infection and are associated with high mortality. Thresholds for surgery during the COVID-19 pandemic should be higher than during normal practice, particularly in men aged 70 years and older. Consideration should be given for postponing non-urgent procedures and promoting non-operative treatment to delay or avoid the need for surgery.     The impact of surgical delay on resectability of colorectal cancer: An international prospective cohort study

    AIM: The SARS-CoV-2 pandemic has provided a unique opportunity to explore the impact of surgical delays on cancer resectability. This study aimed to compare resectability for colorectal cancer patients undergoing delayed versus non-delayed surgery. METHODS: This was an international prospective cohort study of consecutive colorectal cancer patients with a decision for curative surgery (January-April 2020). Surgical delay was defined as an operation taking place more than 4 weeks after treatment decision, in a patient who did not receive neoadjuvant therapy. A subgroup analysis explored the effects of delay in elective patients only. The impact of longer delays was explored in a sensitivity analysis. The primary outcome was complete resection, defined as curative resection with an R0 margin. RESULTS: Overall, 5453 patients from 304 hospitals in 47 countries were included, of whom 6.6% (358/5453) did not receive their planned operation. Of the 4304 operated patients without neoadjuvant therapy, 40.5% (1744/4304) were delayed beyond 4 weeks. Delayed patients were more likely to be older, men, more comorbid, have higher body mass index and have rectal cancer and early stage disease. Delayed patients had higher unadjusted rates of complete resection (93.7% vs. 91.9%, P = 0.032) and lower rates of emergency surgery (4.5% vs. 22.5%, P < 0.001). After adjustment, delay was not associated with a lower rate of complete resection (OR 1.18, 95% CI 0.90-1.55, P = 0.224), which was consistent in elective patients only (OR 0.94, 95% CI 0.69-1.27, P = 0.672). Longer delays were not associated with poorer outcomes. CONCLUSION: One in 15 colorectal cancer patients did not receive their planned operation during the first wave of COVID-19. Surgical delay did not appear to compromise resectability, raising the hypothesis that any reduction in long-term survival attributable to delays is likely to be due to micro-metastatic disease

    Abstract This paper investigates the application of SOA principles for integrating embedded devices features, typically operating in ubiquitous computing environments, with enterprise-level services and business processes. Under this perspective, we present the SAI – Service Application Integration – system as a working example of a message-oriented middleware supporting context-aware application development. A special case study, focused on the monitoring of dangerous goods shipment via maritime transport and involving the gathering, processing and distribution of data collected by heterogeneous components, has been designed to experiment the added value of the proposed integration system in a Web of Things scenario.
